In the same year, she appeared in her second Telugu film, Maha Nandi, with Sumanth and Srihari. Once again, the film failed to satisfy the audience; yet, Anushka's screen presence was greatly appreciated, as were her stunning appearance. The actress had a successful year in 2006, with four films being released.

Her debut film, S. S. Rajamouli's Vikramarkudu, was a commercial triumph. Anushka rose to prominence in Andhra Pradesh after the release of her film Vikramarkudu in 2006. She went on to act in Asthram (2006), the remake of the Hindi film Sarfarosh (1999), before completing her Tamil debut in Rendu (2006). In 2007, she acted in two Telugu films, Lakshyam and Don the former of which performed well at the box office. Anushka starred in six films in 2008, following a moderate year at the box office. Except for Swagatham and Baladoor, all of her 2008 films were successful at the box office.

After succesfully co-starring with numerous South Indian cinema actors, Anushka had her first big solo hit in 2009, appearing in the Telugu horror fantasy film Arundhati. She performed beautifully as two characters in the film. The film was not only a commercial success but also received critical acclaim. Critics applauded her portrayal & she went on win the Nandi Special Jury Prize and Filmfare's Best Actress of the Year. Her next film Billa (2009) was not well appreciated at the box office with her part confined to adding oomph factor. Her second and final release in 2009 Vettaikkaran was also a middling success.

Following a poor finish to 2009 Anushka gave an outstanding performance in the critically praised Telugu film Vedam (2010). Her performance was well received by the public, and she went on to win her second straight Filmfare Best Actress award. However among the films released in 2010, only one Singam was a commercial success. All of her other 2010 releases Panchakshari, Khaleja & Nagavalli were box office disasters.

In 2011 Anushka repeated her role as 'Sarojo' in the Tamil version of Vedam named Vaanam. She was once again commended for her acting with a Sify critic writing: Anushka is excellent as the commercial sexual worker because she is able to carry out a life-like impression". Her second Tamil film Deiva Thirumagal (2011) was a critical and economic success and her acting was once again praised. Anushka also won her initial Tamil Filmfare Award award for her performance. After a two-year hiatus from Telugu cinema she starred in the fantasy film Dhamarukam (2012) albeit her role was more about glamour than performing.

Anushka had a dismal year in 2013, with only one of her releases Mirchi, becoming a commercial success. She did not have much success with her parts in Alex Pandian (2013) & Singam II (2013). Furthermore, for the third time in her career, Anushka played two characters in a film: Varna a high-spirited warrior & Ramya, a doctor in the Tamil romantic fantasy film Irandum Ulagam (2013). She once again surprised the critics by displaying a diverse variety of emotions. She then acted opposite Rajinikanth in the December 2014 film Lingaa, which received mixed reviews.

In 2015 she appeared in the film Yennai Arindhaal which was warmly praised. However, her second movie for the year Baahubali is, went onto to become one of the most successful movies in Indian cinema. In her third film, Rudrama Devi, Anushka played a dual role. Anushka Shetty closed the year 2015 by playing an intriguing portrayal of a plus-sized lady in the film Size Zero. Anushka gained weight for the part in order to portray the character authentically.

Anushka made only a few guest appearances the next year, and her next major appearance came in 2017. She returned to play Kaavya Durai Singham with Si3, the third chapter of the Singam franchise. Her other release this year is Baahubali 2: The Conclusion, a follow-up to one of the most successful films in Indian cinema Baahubali: The Beginning (2015).
